Установлены оптимальные условия хранения венозной крови для проведения ТАБ: не более 4 ч при температуре 18–25 °С. Адаптировано и обосновано преимущество применения форбол-12-миристат-13-ацетата в качестве неспецифического положительного контроля. Исследована экспрессия молекул, относящихся к разным функциональным классам, на неактивированных и активированных базофилах. Показано, что молекулы CD11b, CD13, CD69, CD107a, CD164 и CD300a усиливают экспрессию на базофилах под влиянием антитела к IgE и могут быть использованы в качестве дополнительных маркеров при постановке ТАБ. </p></abstract><trans-abstract xml:lang="en"><p>The basophil activation test (BAT) is a perspective method for allergy diagnosis. However, the optimization and standardization of the method are required. The optimal conditions for the peripheral blood storage for BAT were established: the blood should be stored at room temperature (18–25 °C), and a test should be performed within 4 h after blood collection. The optimal positive control – phorbol 12-myristate 13-acetate was chosen due to effective basophil activation and low cost. The expression of molecules on both unstimulated and activated basophils was studied.
