В исследованиях использован тест по оценке чувствительности рака молочной железы (РМЖ) к химиопрепаратам на основе определения активности клеточной ацетилхолинэстеразы (АХЭ). Сопоставлены результаты влияния химиопрепаратов в культуре и клинического обследования пациенток с РМЖ различных молекулярно-генетических подтипов: люминального А, люминального Б, Her2-позитивного и трижды негативного, или базальноподобного. Установлено, что совпадение результатов по ингибированию активности АХЭ раковых клеток препаратами в условиях in vitro и степени регрессии опухоли пациентки после НПХТ (in vivo) при маммографии составляет 69 %, при мануальном обследовании – 77, при ультразвуковом (УЗИ) – 62 %. Наибольшее совпадение исследуемых показателей получено для самых агрессивных молекулярно-генетических подтипов РМЖ. Разработанные нами молекулярные критерии оценки влияния химиопрепаратов на клетки опухоли в условиях in vitro являются дополнительным прогностическим фактором и доступны к применению в клинических лабораториях при оптимизации тактики лечения пациенток.</p><p> </p></abstract><trans-abstract xml:lang="en"><p>A comparative analysis of the chemotherapy drug effects on the enzymatic activity of breast cancer (BC) cells in a tumor primary culture of BC patients and a clinical response of a cancer tumor to neoadjuvant polychemotherapy (NPHT) with the use of the same drugs were performed. We used our developed test to assess the sensitivity of BC cells to chemotherapy drugs by determining the cell activity of acetylcholinesterase (AChE). The effect of chemotherapeutic drugs in culture and the clinical examination of BC patients of various molecular-genetic subtypes (Luminal A, Luminal B, Her2-positiv and Tripl-negativ or Basal-like) were compared. It is found that the coincidence of the results on the sensitivity of tumor cells in vitro and the degree of the patient’s tumor regression after NPHT averaged for X-ray examination is 69 %, manual – 77 % and US – 62 %. The biggest coincidence of the studied parameters was obtained for the most aggressive molecular-genetic subtypes of BC.
