Цель исследования – лабораторное обоснование оптимальной технологии получения АТК, обеспечивающего максимальные биорегенеративные потенции и минимальный воспалительный ответ при лечении периимплантита.Исследование проводилось на образцах венозной крови, взятой у 30 здоровых доноров (15 мужчин, 15 женщин; возраст – 25–55 лет) без отклонений показателей общего анализа крови. Две группы образцов были получены по разным технологиям: 1-я группа – традиционным методом получения PRP (Platelet-Rich Plasma), 2-я группа – по технологии Cortexil PRP. Полученные АТК изучены с помощью следующих лабораторных методов: атомно-силовая мик­ роскопия (АСМ), подсчет тромбоцитов, лейкоцитов, эритроцитов в каждом слое с использованием автоматического гематологического анализатора, определение концентрации факторов роста и цитокинов – методом ИФА (ELISA).Собранные данные показали преимущество технологии Cortexil PRP, которая обеспечивает оптимальную терапевтическую концентрацию тромбоцитов в PRP, превосходящую традиционную методику PRP по наиболее важным показателям. Так, P/WBC &gt;85 в группе Cortexil – значение, превышающее порог эффективной регенерации (&gt;60); высокий P/R &gt;165 указывает на минимальную эритроцитарную примесь в PRP по технологии Cortexil PRP. АСМ показала, что более 80 % тромбоцитов в Cortexil PRP сохраняли дискоидную форму (признак неактивированного состояния).Качество тромбоцитарного концентрата должно оцениваться по его биологическому профилю, где P/WBC &gt;60 – оптимальная концентрация для подавления воспаления; P/R &gt;120 – для проникновения в кость; &gt;75 % дискоидных тромбоцитов – для контролируемого высвобождения факторов роста; низкий IL-1β/TNF-α – для предотвращения резорбции.</p></abstract><trans-abstract xml:lang="en"><p>This study presents a comparative laboratory investigation of various methods for obtaining autologous platelet concentrates for the treatment of peri-implantitis.The aim of the study was to provide laboratory validation of an optimal technology for producing autologous platelet concentrates that maximize bioregenerative potential and minimize inflammatory response in the treatment of peri-implantitis.The study was conducted on venous blood samples collected from 30 healthy donors (15 men, 15 women, aged 25–55 years) with no abnormalities in complete blood counts. Two groups of samples were obtained using different technologies: Group 1 – traditional PRP extraction, Group 2 – Cortexil PRP technology. The resulting autologous platelet concentrates were analyzed using the following laboratory methods: atomic force microscopy (AFM); platelet, leukocyte, and erythrocyte counts in each layer using an automated hematology analyzer; and determination of growth factor and cytokine concentrations using ELISA.The obtained data demonstrated the advantage of the Cortexil PRP technology, which provides an optimal therapeutic concentration of platelets in PRP, surpassing the traditional PRP technique in the most important parameters. Thus, the P/WBC ratio in the Cortexil group exceeded 85, surpassing the threshold for effective regeneration (&gt;60); a high P/R &gt;165 indicates minimal red blood cell admixture in PRP using the Cortexil PRP technology.
